Abstract

Using pollen from a cold tolerant accession ICC8923, best results for pollen germination and pollen tube growth were obtained in medium containing 2.5% sucrose + 10% PEG + 100 mg boric acid + 300 mg calcium nitrate + 200 mg magnesium sulfate + 100 mg potassium nitrate/litre. The results of pollen germination tests using this medium (PEG medium) and Brewbaker & Kwack (BK) medium are reported. In the BK medium, pollen germination was lower and pollen tube growth was largely confined to the first 60 min with tube abnormalities such as swelling and bursting occurring in 3-h-old cultures. In PEG medium pollen germination was consistently high (<80% after 90 min) and pollen tube length was nearly five times greater than that obtained in the BK medium after 90 min.
