"Cowpea [Vigna unguiculata (L.) Walp.] is an important\r\nfood legume grown in tropical and subtropical regions\r\nof the world, primarily in Sub-Saharan Africa. Despite\r\nthe importance of cowpea, yield on farmers' field is\r\nstill low due to variety of biotic and abiotic stresses that constrain its production. Among the biotic constraints, the parasitic flowering plant, Alectra voge/ii (Benth.) is one of the more formidable limitations to cowpea production in the dry Savannas of West and Central Africa, a region which accounts for over 64 % of world cowpea production. Several control measures\r\nhave been suggested for the control of the parasite.\r\nThese include: cultural practices, application of\r\nethylene chemicals, and host plant resistance. Among\r\nthese control measures, the use of resistance cultivars\r\nappears to be the most attractive option to the\r\nresource poor farmers in sub-Saharan Africa Breeding\r\nresistance cultivars would be facilitated by markerassisted\r\nselection (MAS). The objective of this study\r\nwas to identify molecular markers tightly linked to\r\nAlectra resistance gene that would be useful in MAS\r\nin breeding cowpea for resistance to Alectra vogelii.\r\nF2 population of a single cross, Banjar (susceptible\r\nparent) x B30 I (resistant parent) was screened for\r\nreaction to Alectra using pot culture technique. DNA\r\nwas extracted from parental genotypes and F2 lines\r\nfrom young leaves of plant at 14 days after planting\r\nusing FTA ® PlantSaver cards. 50 SSR cowpea, 40\r\nSSR rice bean and 50 SSR asparagus bean primers,\r\npreviously reported to give amplification products\r\nin cowpea, were used to screen DNA from B30 I\r\nand Banjar for polymorphism. Of the 140 primers\r\nscreened 20 primers were polymorphic between B30 I\r\nand Banjar and these were used in the technique of\r\nBSA performed with DNA bulks of highly resistant\r\nand highly susceptible F2 lines to select those that cosegregate\r\nwith the resistant gene. Two of the markers\r\n(RB 16 from rice bean and CLM0356 from asparagus\r\nbean) were found to be consistently associated with the\r\nresistance gene. The utility of these two markers were\r\nvalidated using 150 F21ines for marker segregation and\r\nassociation analysis. Similarity index (SI) revealed\r\nthat these markers were closely linked (90.23%) with\r\nAlectra resistance gene. Cluster analysis as depicted\r\nby dendogram also showed a tight association (>0.75)\r\nbetween these markers, suggesting that these markers\r\ncan be explored in MAS targeting breeding for Alectra\r\nresistance in cowpea."^^ .
