The plasmid vectors must be carefully chosen and processed to minimize the effort required to identify and characterize recombinants. This chapter provides guidelines for preparation of DNA fragment for cloning, transformation into chemically competent host, and selection of positive clones. The write-up will also describe basic methods used in the cloning of PCR amplified rRNA gene into appropriate vector and followed by sequencing. date: 2013 date_type: published series: Springer Protocols Handbooks publisher: Springer Berlin Heidelberg pagerange: 291-302 pages: 352 refereed: TRUE isbn: 978-3-642-34410-7 book_title: Analyzing Microbes: Manual of Molecular Biology Techniques editors_name: Arora, D K editors_name: Das, S editors_name: Sukumar, M related_url_url: http://scholar.google.co.in/scholar?as_q=%22DNA+Cloning+and+Sequencing%22&as_epq=&as_oq=&as_eq=&as_occt=title&as_sauthors=&as_publication=&as_ylo=&as_yhi=&btnG=&hl=en&as_sdt=0%2C5 related_url_type: pub citation: Kishore Babu, B and Sharma, A and Sudini, H (2013) DNA Cloning and Sequencing.
