"AhMITE1 is an active miniature inverted\r\nrepeat transposable element (MITE) in peanut (Arachis\r\nhypogaea L). Its transpositional activity from a particular\r\n(FST1-linked) site within the peanut genome was checked\r\nusing AhMITE1-specifc PCR, which used a forward primer\r\nannealing to the 50-flanking sequence and a reverse primer\r\nbinding to AhMITE1. It was found that transposition activation\r\nwas induced by stresses such as ethyl methane\r\nsulfonate (EMS), gamma irradiation, environmental conditions,\r\nand tissue culture. Excision and insertion of\r\nAhMITE1 at this particular site among the mutants led to\r\ngross morphological changes resembling alternate subspecies\r\nor botanical types. Analysis of South American landraces\r\nrevealed the presence of AhMITE1 at the site among\r\nmost of the spp. fastigiata types, whereas the element was\r\npredominantly missing from spp. hypogaea types, indicating\r\nits strong association. Four accessions of the primitive\r\nallotetraploid, A. monticola were devoid of AhMITE1\r\nat the site, indicating only recent activation of the element,\r\npossibly because of the ‘‘genomic shock’’ resulting from\r\nhybridization followed by allopolyploidization."^^ .
