"In this study a total of 167 isolates collected\r\nfrom different food materials (68.8% from sorghum and the\r\nremaining from various other food materials) were assayed\r\nby PCR for amplification of the tri 5 gene present in\r\ntrichothecene-producing Fusaria. Amplification of the tri 5\r\nfragment was observed in 45 isolates (39 isolates from\r\nsorghum and 6 isolates from vegetables). Isolates found\r\npositive for presence of the tri 5 gene were classified into\r\ndifferent morphological groups based on their cultural and\r\nconidial characters; 11 of the tri 5 positive isolates from\r\nmoldy grains of sorghum, one from each morphology\r\ngroup were selected for further analyses. Five deoxynivalenol\r\nproducers and three deoxynivalenol and Fusarenon-\r\nX producers were detected by analysing culture\r\nfiltrates of the 11 isolates using GC-MS. One isolate each\r\nwere identified as producers of NIV alone, or NIV along\r\nwith DON or DAS toxins. Identification of these isolates to\r\nthe species level was carried out using spore morphology\r\nand sequence comparison of the translation elongation\r\nfactor 1-alpha (EF-1a) gene against the database as well as\r\nusing phylogenetic analyses. The isolates were identified as\r\nFusarium proliferatum (6), F. nelsonii (2), F. equiseti (1),\r\nF. thapsinum (1) and F. sacchari (1). Amplified Fragment\r\nLength Polymorphism (AFLP) based grouping clustered\r\nthe isolates of same species together. This is the first detailed study of trichothecene production by Fusarium\r\nspp. associated with sorghum grain mold in India and the\r\nidentification of F. nelsonii and F. thapsinum as producers\r\nof trichothecenes."^^ .
