"Seed deterioration is a continuous process and\r\nconservation of germplasm in ex-situ facilities necessitates\r\ntechniques that prolong seed longevity. A combination of\r\n3–7% seed moisture content (mc) and a storage\r\ntemperature below 0°C is suitable for long-term\r\npreservation of orthodox seeds (FAO/IPGRI 1994).\r\nWhen seeds of different crops are dried to low moisture\r\nlevels, there is a decrease in weight and volume and when\r\nlarge seeds dry too rapidly, the outside of the seed loses\r\nmoisture more rapidly than the inside of the seed and it\r\nreduces in volume more quickly leading to cracking of\r\nthe seed coat (Ellis et al. 1985). Internal drying stresses\r\nalso increase susceptibility of seeds to mechanical\r\ninjuries and the dryness of seeds is also critical to the\r\noccurrence of imbibition injury (Ellis et al. 1990).\r\nImbibition injury occurs in standard germination tests of\r\nvery dry seeds, even when the germination medium is of\r\nlow osmotic potential. In addition to legume seeds,\r\nproblems of imbibition injury have been encountered with\r\nforage legumes, cotton and sorghum (Sorghum vulgare)\r\n(Ellis et al. 1985). Imbibition injury to seeds depends on\r\nseveral factors such as seed maturation, age, mc and\r\nstorage temperature (Powell and Matthews 1979, Tully et\r\nal. 1981, Taylor and Prusinski 1990). Imbibition injury to\r\nseeds is a potential problem for genebanks handling\r\ngermplasm samples of very dry seeds especially in\r\nmeeting the requirements for long-term storage. A critical\r\nmc below which a constant proportion of seeds fail to\r\ngerminate as a result of imbibition injury and the\r\nsusceptibility of very dry seeds to imbibition damage has\r\nbeen reported by Ellis (1987) and Ellis et al.
