"Heat shock proteins (Hsps) are a class of\r\nmolecular chaperons which are crucial for protein\r\nfolding, assembly, and translocation in many\r\nnormal cellular processes. They stabilize proteins\r\nand membranes, and can assist in protein\r\nrefolding under stress conditions in plants. Pearl\r\nmillet (Pennisetum glaucum) is highly abiotic\r\nstress tolerant, but its Hsps have not been\r\ncharacterized. In the present study, PgHsp70\r\ngenes were retrieved and gene information\r\nanalyzed in order to characterize their structure,\r\nlocalization and functions. Genome-wide\r\nscreening using the tools of bioinformatics\r\nidentified 18 PgHsp70 genes in the pearl millet\r\ngenome which have been categorized into four\r\nsubfamilies depending on their cellular localization\r\nsuch as endoplasmic reticulum, mitochondria,\r\nchloroplast and cytoplasm. Number of introns\r\nranged from 0-11 in PgHsp70 family genes and\r\nthe genes are located across 1 to 7 chromosomes.\r\nPhylogenetic analysis of Hsp70s revealed that\r\nthey are closely related to Sorghum Hsp70s.\r\nPromoter analysis showed the presence of cisacting elements such as GCN4, HSE, LTR, MBS,\r\nABRE, MYB, and TC Aassociated with abiotic\r\nstress conditions indicating the involvement of\r\nthese genes in the abiotic stress. Under vapour\r\npressure deficit (VPD) conditions, leaf and root\r\ntissues of VPD-sensitive ICMR 1152 line, showed\r\nmild expression and in the presence of high VPD,\r\nVPD-insensitive ICMR1122 PgHsp70 genes\r\nshowed high expression in leaf and root tissues\r\nin comparison with VPD-sensitive line. Gene\r\nPgcHsp70-1 displayed high transcript level under\r\nhigh VPD conditions. These results expand our\r\nhorizon of understanding of the structure and\r\nfunction of Hsp70s, especially under abiotic stress\r\nconditions which can further be validated and\r\nemployed in breeding programs and genetic\r\nengineering."^^ .
