"Pearl millet, Pennisetum glaucumL.), a crop of semi-arid tropics,\r\nhas remarkable tolerance to a wide range of abiotic stresses,\r\nincluding high degree of genetic diversity for drought stress\r\ntolerance. To investigate the molecular mechanisms that possibly\r\naccount for differences in drought tolerance, four recombinant\r\ninbred lines from a high resolution cross (HRC) were\r\nselected for their variability in transpiration rate (Tr) response\r\nto vapour pressure deficit (VPD) conditions. The expression\r\nof PgAQPs in contrasting genotypes varied during the day,\r\ngenerally decreasing in the afternoon regardless of the VPD\r\nconditions. However, under high VPD conditions (4.21kPa),\r\nboth root and leaf tissues of the VPD-insensitive genotypes\r\nhad higher transcript abundance than the VPD-sensitive genotypes.\r\nThree PIP2 subfamily genes (PIP2;1, PIP2;3 and PIP2;6)\r\nparticularly PIP2;6 gene, showed an increase in transcript\r\nabundance under high VPD conditions. Transgenic tobacco\r\nplants constitutively expressing PgPIP2;6 gene were developed\r\nfor functional validation studies in homozygous T2 transgenic\r\ntobacco plants. The transgenic plants showed better tolerance\r\nunder drought stress, VPD and salinity compared to wild type\r\nplants as seen from biochemical, physiological and molecular\r\nstudies. The transgenic plants also showed increased soluble\r\nsugar, membrane stability, reduced electrolyte leakage and\r\nother photosynthetic parameters as compared to the wild type\r\n(WT). Taken together, our studies suggest that, PgPIP2;6gene\r\ncan be deployed to engineer stress tolerant transgenic crops\r\nfor sustained growth and productivity under unfavourable\r\nconditions."^^ .
