"Valencia peanuts are highly susceptible to drought stress.\r\nThe yield reduction due to stress can result in more than 50%\r\nreduction resulting in high aflatoxin contamination. In our\r\npresent study we have compared Valencia-C genotype with\r\na highly drought tolerant runner type peanut C76-16 using\r\ntranscriptome sequencing to investigate tissue-specific gene\r\nexpression and response to abiotic stress. We evaluated the\r\ntwo genotypes under field conditions under irrigated and stress\r\nplots in 4 replications. We found that genotype C76-16 outperformed\r\nyield compared to Valencia-C under drought stress\r\nconditions with only a 30 % reduction in yield while Valencia-C\r\nshowed a higher loss of up to 70%. Each year the yield varied\r\nwidely due to the environmental factors but C76-16 was consistently\r\nranked among the top and suffered a minimal loss of\r\nyield suggesting that it is best suited for the erratic climatic\r\nconditions of southwest U.S. Leave samples were harvested\r\nfrom the control and stress conditions and used for RNA isolation\r\nand sequencing. A total of 340 million paired-end reads\r\nwere generated and were used to find differentially expressed\r\ngenes and transcripts between tolerant and susceptible genotypes.\r\nThe pathways and candidate genes involved in drought\r\ntolerance will provide basic information and expression markers\r\nfor drought tolerance in peanut. We will present our findings\r\non genic content and tissue-specific gene expression and\r\ndiscuss the challenges and opportunities of unifying transcript\r\nsequence data for the peanut community."^^ .
