"Transcriptomic studies are rapidly evolving as a powerful tool\r\nwith next-generation sequencing technology to understand gene\r\nfunctions and molecular mechanisms. RNA sequencing (RNAseq)\r\nprovides a dynamic range for transcript detection and a\r\nbetter quantification of expression levels.With the availability of\r\ngenome sequence in pigeonpea, RNA-seq was used to link the\r\nsequence information to phenotypic traits resulting from specific\r\ndevelopmental processes. In pigeonpea, three-line hybrid\r\nbreeding system is well-established; however, it is technically\r\ndemanding and cumbersome. In order to explore the possibility\r\nof a two-line hybrid breeding system, a coherent transcriptomic\r\napproach supported by physiological and cytological data has\r\nled to the identification of a temperature-sensitive male sterile\r\n(TSMS) line. This line has been characterized for critical (tetrad)\r\nstage and temperature (23°C), and the identification of candidate\r\ngenes involved in abscisic acid signaling for fertility reversion.\r\nFurthermore, a gene expression atlas (CcGEA) has been developed\r\nand transcriptomic profiles generated for studying pod\r\nand seed development with a dataset of 590.84 and 342 million\r\npaired-end reads, respectively in pigeonpea. These data have\r\nbeen analyzed for genes with differential, specific, spatio-temporal\r\nand constitutive expression. In addition, CcGEA identified a\r\ngene network of 28 co-expressed genes, including two regulatory\r\ngenes, a pollen specific SF3 and a sucrose-proton symporter to\r\nbe involved in pollen fertility, which has potential implication in\r\nseed yield improvement. In summary, this study, especially identification\r\nof TSMS and development of CcGEA, will accelerate\r\non-going efforts to enhance genetic gains in pigeonpea."^^ .
