Abstract

Protein contents in C. arietinum seeds were determined by the following rapid procedures: (a) colorimetric estimation of ammonia with phenol-hypochlorite reagents using the Technicon auto-analyzer method, (b) dye-binding capacity method using acid orange-12 dye, and (c) modified biuret methods of Pinckney and Johnson and Craney. Results obtained with the above procedures were compared with (d) the standard microKjeldahl procedure. Results obtained with (a), (b) and (c) were highly correlated with those from (d). The standard error of (c) was high compared with (a) and (b). The possible interference of seed-coat pigments and the effects of flour particle size and time of shaking on the results from (b) and (c) were also studied
