RFLP Analysis of Cytoplasmic-Genic Male-Sterile Lines of Pigeonpea Developed by Interspecific Crosses

Abstract

To study the differences between genetically (GMS) and cytoplasmically male sterile (CMS) lines in pigeon pea (Cajanus cajan), RFLP analysis of total genomic DNA from 3 putative CMS progenies derived from crosses between the wild species C. sericeus and cultivated pigeon pea, and 7 C. cajan accessions (including 2 GMS lines) was performed. After the initial cross, C. sericeus EC121208 × C. cajan ICPX880227, transfer of the nuclear genome of pigeonpea into C. sericeus cytoplasm was achieved by 5-6 subsequent genomic transfer stages using different male pigeon pea parents. Hybridization patterns clearly distinguished between GMS lines ICPL87 and MS Prabhat, CMS lines produced by multiple gene transfer and pigeon pea lines, although there were also differences in the restriction fragment patterns among the various pigeon pea cultivars, and between MS Prabhat and ICPL87. Results suggest that crosses between C. sericeus and cultivated pigeonpea produced male sterile lines having mitochondria of the latter.
