Abstract

In peanut, functional genes associated with various metabolic pathways, including carbon fixation, aminoacyl-tRNA biosynthesis, pentose phosphate pathways, starch and sucrose metabolism, flavonoid biosynthesis, hormone signalling, and photoperiod-dependent pathways, were identified across developmental stages of different tissues and organs. Gene expression related to key quality traits, such as fatty acid content, fresh seed dormancy, and crack-free seed coat, was also demonstrated. Additionally, genes responsive to abiotic and biotic stresses were identified, and the mechanisms underlying resistance reactions were elucidated. Through the creation of functional variants via mutation, several improved peanut cultivars were released. Post-transcriptional regulation of functional genes was explored using RNA interference (RNAi), small RNAs (sRNAs), and virus-induced gene silencing (VIGS). Detailed proteomic studies focused on the peanut peg, pod, seed, allergens, and stress-resistance mechanisms. Proteomics was employed to decipher the genes and pathways involved in the Aspergillus–peanut interaction and to identify key factors involved in oleic acid biosynthesis. Several genes were expressed in planta and functionally validated through the development of high-frequency regeneration and transformation protocols.
